New Investigators must meet with the Core team to discuss their project before sample submission can be arranged. Investigators are expected to load samples onto plates following the instructions below.
*Investigators can submit individual samples in tubes and request sample plate loading by the Core for an additional fee.
A. Preparation for Olink run
- Email Core staff (AffinityProteomics@bch.harvard.edu) to discuss and arrange your study.
- Prepare your sample manifest using the provided template. Please modify the manifest file name to include date, institute, user last name, and specified panel (i.e. T48_01012027_BCH_Lee_humancytokine_manifest). Fill out the highlighted cells with your sample ID. Data output will be aligned to the sample ID provided in the manifest.
- Confirm plan with Core to transfer Investigator-plated thawed samples on the scheduled date of sample submission, or transfer Investigator-plated frozen samples prior to scheduled date.
- Arrange date and time for sample transfer with Core staff (Monday to Thursday, 9 AM to 1 PM).
B. Sample plating by Investigators
- Print out your sample manifest. Use this as a reference while you plate the samples.
- Prepare a biosafety cabinet, sterile pipette tips, and a polypropylene 96-well PCR plate. Please note: we will only accept Investigator-plated samples delivered in polypropylene PCR plates. We will not accept samples delivered in flat-bottom, round-bottom, or V-bottom tissue culture plates. The Core can provide a PCR plate for sample plating upon request.
- Pipette at least 10 µL of each sample into the PCR plate according to the following map. Observe sterile technique during plating to avoid contamination. After pipetting, seal the plate tightly with an aluminum adhesive film (ensure the film is compatible for cold storage to -80C).
For Target-48 and Olink Flex panels, use the following arrangement for plate loading. Columns 6 to 12 should remain empty.
